UMAP embedding of genetic dependencies of repair outcomes at Cas9-induced DSBs

The upper left panel summarizes the genetic dependencies of individual repair outcomes from screens at four different Cas9 target sites, as in Figure 3G here.
Outcomes are initially colored by the effect of a POLQ-targeting CRISPRi sgRNA on their frequencies.
Use the menu to the right to select different CRISRPi sgRNAs, or the tabs above to color outcomes by other properties.
The bottom panel shows the correlation in outcome frequency redistribution between the currently selected CRISPRi sgRNA and all other sgRNAs.
Mouse over points in the bottom panel to compare the effects of other sgRNAs.

Selecting CRISRPi sgRNAs

Selecting a CRISPRi sgRNA in the dropdown menu colors outcomes by the effect of that sgRNA and updates the bottom plot to show correlations with the selected sgRNA.
The top 10 sgRNAs with highest correlation to the selected sgRNA are labelled.
Alternatively, with focus on the menu, type an sgRNA identity and press Enter, or use the up and down arrow keys to cycle through sgRNAs.

Visualizing other outcome properties

Click on tabs above the main plot to see how other properties of outcomes are distributed across the UMAP.

Comparing CRISPRi sgRNAs

To compare the effects of CRISPRi sgRNAs, mouse over an sgRNA's point in the bottom plot to pop up a second copy of the outcome UMAP colored by changes in frequency for this second sgRNA.
